چکیده

Introduction Mevalonate kinase deficiency (MKD) has a wide spectrum and severity of clinical manifestation. Patients with mutations in MVK gene leading to complete lack of the enzyme, suffer from the most severe form of disease, also known as mevalonic acydosis, whereas defects with preserved, but insufficient enzyme activity present with autoinflammatory syndrome , also known as hiperIgD syndrome (HIDS). Both diagnosis and treatment of MKD is a great challenge for clinicians. Hematopoietic stem cell transplantation (HSCT) is the only available therapy that allows delivery to the tissues of the missing enzyme produced by healthy donor hematopoietic cells.

Long-term outcome of a successful cord blood stem cell transplant in mevalonate kinase deficiency.

Mevalonate kinase deficiency (MKD) is a rare autosomal recessive inborn error of metabolism with an autoinflammatory phenotype that may be expressed as a spectrum of disease phenotypes, from those with prevailing autoinflammatory syndrome and variable response to anti-inflammatory therapies, to mevalonic aciduria, which is associated with dysmorphic features, severe neurologic involvement, and ...

12.2 Mevalonate kinase deficiency: Impaired isoprenoid synthesis induces IL-1beta production via activation of Rac1

Mevalonate kinase deficiency is an autosomal recessive disorder characterized by recurring episodes of fever and inflammation. Peripheral blood mononuclear cells from mevalonate kinase deficiency patients secrete high levels of IL-1β when stimulated with lipopolysaccharide (LPS) due to the presence of hyperactive caspase-1. The molecular mechanism of mevalonate kinase deficiency-induced caspase...

Diagnostic value of urinary mevalonic acid excretion in mevalonate kinase deficiency (MKD)

Introduction Mevalonate kinase deficiency (MKD) is a rare hereditary autoinflammatory syndrome, characterized by recurrent fever episodes with gastrointestinal complaints, rash and arthralgia. The deficient mevalonate kinase activity leads to elevated mevalonic acid, which is excreted in the urine. Therefore, an elevated mevalonic acid excretion is suggestive of MKD. However, the diagnostic val...

Perinatal manifestation of mevalonate kinase deficiency and efficacy of anakinra

BACKGROUND Mevalonate kinase deficiency is a metabolic autoinflammatory syndrome caused by mutations in the MVK gene, mevalonate kinase, the key enzyme in the non-sterol isoprenoid biosynthesis pathway. Two phenotypes of mevalonate kinase deficiency are known based on the level of enzymatic deficiency, mevalonic aciduria and hyperimmunoglobulinemia D syndrome, but a wide spectrum of intermediat...

Mevalonate kinase deficiency: current perspectives

Mevalonate kinase deficiency (MKD) is a recessively inherited autoinflammatory disorder with a spectrum of manifestations, including the well-defined clinical phenotypes of hyperimmunoglobulinemia D and periodic fever syndrome and mevalonic aciduria. Patients with MKD have recurrent attacks of hyperinflammation associated with fever, abdominal pain, arthralgias, and mucocutaneous lesions, and m...
